The Cross River State Police Command has appealed to members of the public for information that could help identify the victim and assist investigators in establishing what happened.
Police said preliminary investigations showed that the deceased lodged at the hotel alongside a male guest whose whereabouts were still being investigated.
The authorities have also stressed that individuals being held in connection with investigations should not be regarded as guilty unless established by the appropriate legal process.
Other alleged killings have previously been reported in parts of Cross River, including Yala, Ogoja, Ikom and areas around Calabar.
